Efrayim Dov Lando was born on 5 April 1930 in Zgierz, Poland, to Tuvia Yosef and Devorah Lando. On his father's side he descends from the Strykow Hasidic dynasty. In March 1935 the family moved to the Land of Israel and settled in Rehovot.

He studied at the Yishuv HaHadash Yeshiva, then at Ponevezh Yeshiva for several years and later at Hebron Yeshiva for one year. He considers the Chazon Ish, Avrohom Yeshaya Karelitz, his main teacher. Although his family was Hasidic, he joined the Lithuanian stream from his youth.

He married Adina, a granddaughter of Slabodka rosh yeshiva Isaac Sher, settled in Bnei Brak and taught at the Slabodka Yeshiva. He later became one of its heads, alongside rabbis including Moshe Hillel Hirsch. In 1988 he opposed Degel HaTorah running separately from Agudat Yisrael in local elections.

He joined the presidium of the Board of Yeshivas in 2014. After the death of Gershon Edelstein he became co-chairman of the Moetzes Gedolei HaTorah with Hirsch. In 2023 to 2025 he made statements opposing army service for yeshiva students and criticizing Zionism, which drew condemnation from politicians including Naftali Bennett and Gadi Eisenkot.
